Basic scientists studying pain and targets for analgesia are continually exploring unique mechanisms to further our knowledge of such systems. The ultimate goal of these efforts is the development of novel therapeutics to help those suffering from chronic pain. As new molecular mechanisms are uncovered, hopes arise that compounds will be developed for these new pain targets. However, in the excitement over such new and potentially efficacious targets, their possible side effects may be overlooked. The design of antagonists for the TRPV1 channel is being pursued by many pharmaceutical companies and the clinical relevance of such antagonists will soon be elucidated. The discovery by basic scientists of alternative TRP channels as well as how such a family of channels may be regulated by other proteins leads one to believe that these compounds will be therapeutically developed for pain relief.
This symposium brings together scientists who are actively involved in the development of drug(s) for a molecular target that was discovered and explored by basic scientists. The discovery of TRPV1 and subsequent developement as a clinical target make it an excellent model for making a transition from benchside to bedside.
Prisca Honore, Ph.D. (Abott Pharmaceuticals) will describe the development of a TRPV1 antagonist for pain. Narender Gavva, Ph.D. (Amgen Pharmaceuticals) will discuss how testing such antagonists for the TRPV1 channel has resulted in difficulties in clinical trials and how the limiting side effects could have been detected at the basic science level. Gil Wong, M.D. will discuss the use therapeutic use of TRPV1 agonists instead of antagonists and finally an undetermined individual will discuss other TRP channels as possible therapeutic targets.
Brief presentations by each speaker will be followed by an extended discussion. Members of the APS basic science research community will be highly encouraged to participate.
Drs. Vanderah and Stone will serve as moderators facilitating discussion.
